Assuming the chip will be programmed off line, can the 'serial in' pin be used as general input pin?

If used as a truly general purpose input, the hardware should ensure that the Serial In is not allowed to be taken high until the PICAXE has executed a DISCONNECT command after starting. Otherwise the PICAXE may not operate as expected.

Exactly as Hippy has said, but a nominal 10k pull down resistor from C.5 (seriin) to ground will replace the download circuit.
